The purpose of this study is to evaluate the effects of fluoride administered
systemically during enamel formation on developing enamel experimentally. Subcutaneous
injections of sodium fluoride solution were given to the Sprague-Dawley rat pups at
various days of age and with various daily doses. At 14 days of age, the rats were
sacrificed, the mandibular first molars were extracted, and the enamel was sampled
using acid-etching biopsy.
The fluoride concentration and the solubility of enamel were measured using fluoride
ion electrode, atomic absorption spectrophotometer and wavelength dispersive X-ray
spectrometer, and the structure of enamel was observed using scanning electron
microscope.
The following results were obtained;
1. The fluoride concentration of enamel was increased and the solubility decreased
following systemic administration of fluoride during enamel formation. There was a
negative correlationship between the fluoride concentration and the solubility of enamel.
(correlation coefficient; -0.7829)
2. There was a trend showing that the earlier the fluoride administration was started,
the longer continued and the more fluoride dose was administered, the more the fluoride
concentration of enamel was increased and the solubility decreased. If the fluoride
administration was done once before eruption, the amount of increase in fluoride
concentration and decrease in solubility was most in the group which was administered
on 5 days of age.
3. The fluoride concentration of enamel in the F-injected group was higher than that
in the control group at all depths of enamel (p<0.01),
4. The fluoride concentration of enamel was lower at 20 ¥ìm deep layer from enamel
surface than the other layers of enamel both in the F-injected group and in the control
group(p<0.05).
5 The structural change of enamel was observed in the F-injected group.
